Purpose: The overall objective of the project is to use gold-nanoparticles functionalized with curcumin (cur-AuNPs) for targeted radiotherapy of prostate cancer. The idea is to employ a multimodal treatment via the chemopreventive properties of curcumin, radiosensitization, dose enhancement from external radiation, and enhancement of tumour imaging. In this study, we focus on the effects of synthesis parameters on the particle-size distribution of cur-AuNPs along with their colloidal stability over time.
Methods: Cur-AuNPs were synthesized through dropwise addition of 200µL of 100mM HAuCl₄.3H₂O to 2mM curcumin solution (prepared in 1,3 or 5 mM sodium hydroxide) held in an ultrasonic water-bath at 21,32,or 44 °C. The total yield of cur-AuNPs at different temperatures was measured using inductively-coupled-plasma-optical-emission spectroscope (ICP-OES, Teledyne Leeman Labs) technique. The stability of cur-AuNPs, synthesized at 44 °C, was analyzed (using Malvern Zetasizer Nano-ZS) 1,11,22,and 29 day(s) post synthesis.
Results: The formation of the cur-AuNPs was first confirmed with the appearance of a ruby-red color. The peak intensity (average particle-size) of cur-AuNPs, synthesized with 1,3,and 5mM sodium hydroxide (NaOH), was found to be (i)96.3±4 nm, (ii)47±1.3 and 3±0.3 nm, (iii)22.6±0.1 nm, respectively. The particle-size distributions of all three samples measured the same up to 29 days post synthesis.The Zetasizer measurements of cur-AuNPs, synthesized at 21,32,and 44 °C, was found to be 23.97±0.87, 21.9±1.01, and 22.6±0.5 nm, respectively, with relatively good polydispersity index (~0.15). These measurements showed single peak intensity, indicative of uniform dispersion of particles. The gold concentration, obtained via ICP-OES, gradually increased with increase in temperature of the water bath; 399.4,423.8,and 460 µg/mL for cur-AuNPs synthesized at 21,32,and 44 °C, respectively.
Conclusion: Different size-distributions of relatively stable cur-AuNPs can be synthesized by varying the molarity of NaOH solution. Although varying the reaction temperature did not affect the particle-size distribution, the overall yield of cur-AuNPs increased with temperature.
