Purpose: Osteoporosis is a systemic skeletal disease feathered by low bone mass density and microarchitectural changes of bone tissue. The purpose of our study is to investigate the feasibility of characterizing bone microstructure by analyzing the frequency spectrum of xray acoustic computed tomography (XACT) imaging of the bone samples.

Methods: K-wave simulation of XACT signal were performed on trabecular bone simulations and micro-CT scans the images between osteoporosis and healthy bone models in mice.

Results: In our study, the simulation based on osteoporotic models have proved that there are thinner trabecular thicknesses in osteoporotic bone samples in comparison with normal ones. Additionally, the results of quasi-linear photoacoustic spectra fitted by linear regression demonstrated that much higher spectral parameter slope were relative with osteoporosis. Experiments on osteoporotic mice tibia with different mineral content was conducted. The finding from the experiment was in good agreement with the simulation, demonstrating that the frequency-domain analysis of XACT signals can provide an objective assessment of bone microstructure and deterioration.

Conclusion: Considering that XACT imaging is inherently a dual modality imaging technique capable of providing acoustic imaging information in addition to the transmission xray image. Therefore, this new bone mass evaluation method based on K-wave simulation and XACT spectral analysis holds potential for clinical management of osteoporosis and other bone diseases.
