Metal-Organic Framework (MOF) Based Nanomaterials as Radiosensitizer and MRI Contrast Agent

Purpose: To introduce a novel multifunctional nanomaterial based on Fe3O4 core and high-Z metal-organic framework (MOF). The the radiosensitization ability and drug loading capacity of this nanomaterials were investigated.

Methods: A layer-by-layer growth method was employed for the synthesis of UiO-66 (Hf) shell on Fe3O4 nanoparticles core, the shape was characterized by Transmission Electron Microscope (TEM)and a Monte Carlo simulation based on the TEM image was performed to investigate the radiosensitization of these nanoparticle. To verify the drug loading capacity of these nanoparticle, drug release experiment was conducted by monitoring the UV-vis absorption spectra of the released ibuprofen at different time points.

Results: The Fe3O4 nanoparticles are clearly coated by the shell of MOF, the size of Fe3O4 core is around 170 nm and the thickness of shell of Metal Organic Framework is around 20 nm. Drug loading experiment shows that this nanomaterial has great drug loading capacity. MC simulation also demonstrates Fe3O4@MOF nanomaterial have great radiosensitization performance and as the thickness of Hf-based MOF shell increases, the DER increases accordingly.

Conclusion: The multifunctional nanomaterial has great potential to be used as radiosensitizer and drug carrier. And due to the Fe3O4 magnetic core, they could also be used as MRI contrast agent. Future study would focused on cytotoxicity and therapy efficiency of this nanomateiral by animal experiments.
